Multiple epitopes of HCV proteins (Core, NS3, NS4, NS5) are bound to microtiter wells. Test samples react with recombinant proteins, and bound human IgGs react with anti-human IgG peroxidase conjugate and chromogenic substrate. Positive reactions produce a blue color that is quantified photometrically.

1. Equilibrate reagents to room temperature
2. Dispense 100 µL of specimen diluent into each test well
3. Add 100 µL of positive and negative controls in duplicate
4. Add 10 µL of test samples in duplicate and mix
5. Incubate for 30 minutes at 37°C
6. Wash wells 5 times
7. Add 100 µL of Enzyme Conjugate and mix gently
8. Incubate for 20 minutes at 37°C
9. Wash wells 5 times
10. Add 50 µL Substrate Solution A and 50 µL Substrate Solution B to each well, mix and incubate for 10 minutes
11. Add 50 µL Stop Solution and read OD values at 450 nm/630 nm
